My first thought is Vagabond. Ive been reading that since I was a 10th grader and its still not animated what gives?!

Another series I enjoyed was Alive The final evolution. I really dug the characters even if it wasnt the most otiginal series out there. Was really bummed it was cut short due to the creator passing on. While the ending was rushed because of his declining health he did give us an ending before that...I felt gratitude for that. A bit of his emotions must have bled unto the final pages. Anywag itd be nice if an anime studio would pick this series up and do something creative with it.

Oh and I hear 20th century boys was a good manga? Its been years but maybe theres hope? Naoki urasawa gave us monster so how anout this?

Vagabond will never get an anime adaptation, apparently. But I prefer it to stay in the manga format. It's much deeper and portrays better author's intentions. The anime might just ruin the whole series' concept, if done wrong. Though, I might be okay with a short OVA.

GTO needs a seqel.
Ahiru no Sora needs an anime adaptation too, but it's not that popular. I still hope for it though.
20th CB also needs one.

Vagabond,Vinland Saga and Historie all need anime.

How about Holyland.Can't say I enjoyed it that much but I think it needs an anime adaptation.Although I'm very much skeptical in that matter after seeing how shitty adaptations have ruined the original manga versions for most.

Going with a visual novel here, but "Saya no Uta" made into a series of movies/OVAs in the likes of "Hellsing Ultimate" or "Kara no Kyoukai: The Garden of Sinners".

Hoshi no Samidare. Fast, lively, colorful, brimming with energy, and cleverly poignant. There's little reason to not adapt this manga.

Other than that, I'd love to see an adaptation of Eden: It's an Endless World. But it'd need a crazy budget and the dude who directed GitS: SAC to even stand a chance of being equal to the manga.

I would be interested on longer/more faithful adaptations of Battle Angel Alita and Akira. I haven't read their mangas, but they are supposed to be major works and it seems their movie adaptations both suffered the problems of compressing.

And Yotsuba. The day it gets an anime I will open a bottle of champagne or something.

Hiyokoi needs a proper adaptation.

Nukoduke could also do with 12 or so shorts like Danna ga Nani wo Itteiru ka Wakaranai Ken and Himegoto.

Rewrite will never be done well unless it gets more than 24 episodes and a decent studio working on it. That game had far too much to cover.

Hibi Chouchou because the art is really beautiful. I want to see those cute characters move :3
Tower of God because it's epic and Cheese in the Trap because it's one of the best romance stories I've ever read. Both are Korean webtoons though so it's almost impossible to see them getting anime adaptation at this point.

Dengeki Daisy. It's the best romance manga I have ever read.
Shin Angyo Onshi. I know a 1 episode animation exists, but it was terrible and, of course too short for a manga almost always included in the top50 mangas.
Hoshi no Samidare. It's like Beelzebub, but actually better.
World Embryo. It's similar to Tokyo Ghoul, but not as dark.

Alive is pretty underread around here, tbh. I think it's decent and would definitely do well as an anime! It was made by Noragami's mangaka, btw.

Definitely 20th CB.
