Now we all now there are a lot of stock location for an anime to take place in. The most notable of which being high school. However what anime has the most unique setting that it take place.

My pick would be Magi. I can't think of a single other anime that take place in an ancient middle eastern setting. Part of makes Magi standout is its setting and the way the characters dress. I wish a lot more anime would take place other countries for varieties sake.

Ahh, you beat me to it with Magi. I've been saying for years how superb the setting of that show was, along with the characters' attire.

My next answer may seem like a surprise, but, One Piece is very unique with its setting, as not only does the scenery change very frequently, but, the backdrop of pirates seems to be ultra rare for Anime. I can count the amount of pirate Anime I've seen/heard of on 1 finger... that finger being dedicated to One Piece.

one piece the amount of places the strawhats travel to and the unique way they dress to fit their environment for example in wano.

Morioh from jojo part 4 or the main setting for part 7 are the first that comes to my mind.

Morioh is a really memorable and unique location even in jojo standards in my opinion. The whole part 7 setting and concept is one that i've never really seen be used anywhere else either.

I don't like to use "the most" because it's not like I've watched every single anime, but I've at least never heard of any one with the same "Halloween" setting Soul Eater has, even though it's also about high school.

The main setting of Galaxy Express 999 is a train.
Also, not absolutely unique, but the underwater setting of Nagi no Asu kara is quite original.
And then there is Cells at Work, inside a human body.

One Piece definitely. Maybe HunterxHunter can get close but yeah One Piece has some crazy places in it's world setting. It's geography is like a 'fuck you' to physics.

Kujira no kora wa sajou ni utau, of the anime I've seen. Its amazing setting is its saving grace honestly.

I agree with a lot of what was mentioned already, but I don’t think I’ve seen someone mention Michiko and Hatchin yet. I thought it was pretty cool to see an anime take place in Brazil (at least I think it was Brazil it’s been a while since Iv’e seen it)

Tower of God, I can finally mention it in anime discussions, but the great setting and atmosphere is 100 times better in the manga, but the overall world and setting are outstanding in quality and uniqueness.

After Magi, the next unique setting that comes to mind is from Ancient Magus Bride, English forest settings is not something we see often.

Houseki No Kuni, Shinsekai Yori, Dorohedoro.
These three absolutely nailed it with their unique and out of the box premise. These shows definitely brought something new to the table for me.

I have to go with Re zero.The characters are so well written and it has one of the best world building.
Till now I have seen 3 isekais and the most unique Isekai I have seen will be Re zero.

One Piece, Oda didn´t created a Manga Oda created a world and in doing so he made the best manga of all time.

Other one would be Made in Abyss.

Attack on Titan. Because it never needed dumps of dialogues and exposition to prove the credibility of its world and even though giving a very classic old age historical vibe to its atmosphere , it stands out regardless especially in the latter chapters. The expansion doesn't just perpetuates just for the sake of it without limit and reason like a lot of shows aforementioned . Its never expands in adventures manner but like revealing manner. All the world buildups have significance and consequence to the entire thing as whole which just makes the existence of this world stand out and one of a kind.

For me it would be A Certain Magical Index, world where magic and science cross paths. I have never seen world this refined where elements of both scifi and fantasy works so well. And the beauty which novels bring later in the story is just unforgettable.

Girls' last tour I mean its the most realistic dystopian setting I have ever seen. The characters just live and do nothing about it, they realize that that's just their life now and they just have to live with it. Oh and the visuals are amazing

Well, a lot of the ones I was going to mention were already posted here, so I'll say Last Exile and Gankutsuou had pretty unique settings, though the latter isn't as developed as the former in that department.

Made in Abyss. Concept of "another world" inside a giant hole on the ground and the consequence of seeking that place is what i think my most unique setting from other anime that i watched.

When i watch this anime, i felt my curiosity is reaching to the top, wondering if the scenario really happen in this world..

Apart from the ones mentioned, I would go with
Dagashi Kashi - it's all about the sweets
Death Parade - the afterlife games to decide which path the dead may follow
Dimension W - I don't recall ever watching an anime that explores the existence of a 4th dimension
Dr. Stone - going back to the stone age and rebuild the world as we know
Fune wo Amu - apparently, creating a dictionary has its charms
High Score Girl - revolving around the videogames world and its evolution
Kore was zombie desu ka? - a Magical Girl Zombie Guy?

I recommend Arslan Senki. It has similar setting albeit very simple story.

My best bet would be One Piece. There is just no limit to whatever kind of islands, locations, characters, and even culture in this world. It has a plethora of its own kind, its lore is something I would not even imagine even if I'm influenced with drugs. Yet, here they are. These are the most mystifying for me:

Neo Venezia from Aria is literally just Venice, Italy but it's still one of the most beautiful worlds I've seen in an anime.

For me it's Juuni Kokuki. At first glance the setting might not look all that unique but the world's politics and mythology is explored in great detail and it's not just useless info dumps but plays a vital role in how the the story develops.
