Short-tempered females are abundant across all genres of anime, particularly harem, but I have not seen one take the lead role in any anime. Can you tell me some examples of anime with a tsundere lead? And does it/would it work well?

Well, do you mean solo lead? Because if not, there are plenty of female lead examples such the titular Shana of Shakugan no Shana.

The problem with having a female tsundere as a solo lead is that the target of their anger/affection would have to be towards a single male (usually) character; surely that would implement the male character as the lead role, if not deuteragonist.

Basically, I think it's difficult to have a tsundere solo lead. I'm sure there are some examples of it having been done, but not entirely sure if I've watched any.

Haruhi is a tsundere lead. Maybe just tsun without dere. But it is rare cause tsundere is a sexual fantasy of girls in real life. IRL otakus get rejected by girls and get treated harshly by them. In their escapism entertainment they dream that those bitches actually just fake their hostility to hide their affection. This only works if the tsundere is in a romantic relationship with mc-kun. As an MC of an Anime this wouldn't work.

It depends on what type of tsundere you're looking at.

Victorique from Gosick is a tsundere, I believe, but you'll have to ask @zefyris on that since they're more knowledgeable on this than I am.
She's also far better than the vast majority of them, whilst being the lead character.
It's an entirely different kind of flying, altogether!

I think Hinamori Amu from Shugo Chara! is a bit of a tsundere and she's the main character.

Mikoto Misaka from Railgun is considered to be a tsundere.

Kaichou wa Maid Sama. She is a tsundere and the story is from her perspective. While a lot of others named above are tsunderes and main characters, the series does not focus from their perspective.

Victorique is a main character but the protagonist (if I understand what the OP want by "lead character here,because AFAIK in english lead character = protagonist ?) is Kujou, not her.

Same for Zero no Tsukaima,shakugan no Shana and so on. they are main characters and the main girls, but not the lead character.

Haruhi isn't even a tsundere. To confuse Haruhi with a tsundere, you must have a reaaaally low understanding of that concept. So obviously anything else y usaid in your post is to be considered with this in mind. And hoooh, what do we see. Affirmations that reeks of ignorance and prejudice. Where did you pull that idea from? Wait. Don't answer that part, I don't want to know.

Taiga isn't even a Tsundere to begin with. That's going to be difficult to make her a "perfect example". Basically, Taiga is always violent (except with her love interest), but honest with her feelings.
Tsundere are defined by their inability to be honest with their feelings.

I should really start to save those explanations to avoid typng them again and again lol...

Because "Tsundere" is a japanese slang referring to being unable to be honest with his/her dere feelings toward someone/a group of person, and therefore that character keep being "harsh " toward those persons to hide those feelings due to their embarrassment.
It's basically an exaggerated version of the "young boy who has a crush on a girl and keep pestering her/being harsh toward her because he cannot be honest".
Aisaka Taiga is the exact contrary of this.
-She is violent toward anyone that she isn't in love with.
-She's very honest about her feelings. In fact she even confessed to her love target right in the first volume (how many non-tsudnere characters are that quick and honest actually?). And then when she heard from her second love target that it's okay to go out with him, she jumps immediately in his arms and say she won't ever let him go. Furthermore, she stopped being harsh toward that second love target BEFORE realizing she was in love with him. So yeah, she's honest about her feelings, harsh with others but not toward her love target.

The reason peoples confuse her with a tsundere are multiple.
1) She's voiced by the tsundere queen. Doesn't help at all. But as a reminder, lots of non-tsundere characters are voiced by her too).
2)She has a violent personality and fall in love. Peoples on English boards easily confuses this as being tsundere, because they don't understand the slang's meaning. Being violent isn't being a tsundere. And obviously ANYONE can fall in love. This doesn't make those who are violent tsundere. Being violent is a CONSEQUENCE of their embarrassment when they're with their love target, IE being violent (with words or physically) is not the cause of calling her a tsundere, it is a consequence of the character being a tsundere (and not systematic to boot).
Peoples on English boards needs to understand already that if a girl kick you in the balls because she thought you were trying to molest her, and then 3 months later falls in love with you, this won't make her a tsundere. Tsundere is a PERSONALITY trait (and usually an exaggerated one). Not a succession of event. Taiga has a violent personality. But not a tsundere personality.

3) When peoples on English boards explain to others what a tsundere is, they have a tendency to take Taiga as an example (among others). OK....they are not the most likable, but they are intriguing, if only for their bipolar attitude....

Actually, growing up, one of my favorite shows was Nickelodeon's Hey Arnold....I didn't know it back then, but it had one of the BEST tsunderes to ever be animated...
