Usually (not always) when virginity is emphasized in an anime it's targeting a certain audience.
$$$

It also happens in eroge, though in that medium the intention is much more blatant, and we all know what the majority of those titles are designed for.

i.e.
The cast of K-On would not have given certain viewers as big of a hard-on had they not been portrayed as highly exaggerated, childlike virgins.
That would have meant less popularity and sales.

I guess it has something to do with the Gap. For example because of the way a person acts or who she is hanging around people would assume she is experienced.

When discovering she is a Virgin, the image of her sleeping around disappears.

Aside from a few jokes along the lines of 'OMG character X is so pure!' (which I wouldn't really take seriously), I've never seen virginity being emphasized in any anime. Also, even if it would be, I don't see the issue. Most characters are around highschool age (15-18? feel free to correct me) anyway, and taking that as well as the cultural differences of Japan and western countries into account, does it really make for such a great surprise that a lot of characters are virgins? That being said, often times it is never stated whether or not a character has had a relationship in the past, so it's not like you can always tell.

If a man insist his wife should work outside or stay home he would be bashed as if he were criminal because women must have rights to choose work outside or not
Most men don't have rights to choose whether work or not

It's honestly some weird fetish/expectation that otaku have when viewing female characters. Used goods are no good for them. The prevalence of such an idea in anime is due to studios pandering to that otaku audience.

Well I can understand the reasons and maybe people will think I'm crazy but it has actually influenced me to be scared of sex. I'm planning to stay virgin because I'm afraid I'll lose my dignity. Anime has actually played a big role in it, unconciously.
