Will there be romance between Igarashi and Takeda?

Yes she's a VA but she's still an adult and she is speaking like an adult, you are probably just too used to children being voiced by adults.

Beyond the tonal aspect, which is largely irrelevant, it is the vocabulary that's used and the way the speech patterns convey emotion that are most tellingly adult.

At the root of this is just you seeing a small woman and equating that to child and extrapolating to loli, which is a bit dumb.

Megumin is 14 (I think) in lots of societies she would be a young adult

How the VA speaks in real life is irrelevant, she is not voicing Igarashi as a child, the show does not show Igarashi doing childish things

This show is a comedy. Don't take things too seriously. If you get repulsed by main freaking characters, then find yourself a different show. Romance is obviously inevitable here. This isn't a drama show, so there won't be no teary "let's be friends", or "I see you only as a father/daughter" ending with MCs parting ways and moving on with their lives. In other words, there won't be no father/daughter relationship. I haven't read the source, but for someone who has seen/read other COMEDY-romance shows, the outcome should be OBVIOUS. If you're planning to start watching this show with such party pooper POV, you will only have a bad day; rant and rant and rant. I'd say, spare yourself the trouble...
