That blood wasn't DIO's, it was Joseph's blood, DIO sucked his blood and with the transfusion they gave back that blood to Joseph. But even if that could transform Joseph into a vampire he has Hamon so he is protectrd

There’s a few reasons
1) Joseph has hamon
2) he wouldn’t turn into a vampire unless dio gave him his vampiric essence directly. He says so to johnathan in part 1
3) it’s Joseph’s blood just being returned to his body
