SPOILERS IF YOU HAVE NOT SEEN THROUGH THRILLER BARK
I believe that Usopp is not and has never been a coward. Starting in Syrup Village, when Usopp learned that pirates were going to attack the village he didn't run away. He was going to fight the pirates on his own without anyone's help. And yes, he was scared in this moment BUT that was justified, he was like a 16 or 17 year old kid who had never been in a fight before and was about to stand up to an entire crew of pirates on his own. And during the fight, even though Luffy and Zoro did most of the work, he still held his ground and even took out Django with Zoro's help. What I believe happened is that he got the idea that Luffy and the others helped him because he was scared, and started to lie to himself about being a coward. Nothing Usopp did during the Syrup Village arc made him out to be a coward.
Next, in Arlong Park, he ended up in the position where he had to fight on his own. When his fight started, he ran away in hopes that someone would come to help hm, but after a while realized that no one was coming. He realized that pretending to be a coward with no one around was useless, so he started to act like himself again and absolutely demolished his opponent.
After that, when he was in Alabasta, he and Chopper had to fight a pair of enemies. This time was slightly different, as Chopper never believed any of his lies about being a coward, Chopper looked up to Usopp as a great fighter because Usopp said things like that when they weren't in danger and Chopper had never seen Usopp fight before then. Usopp noticed this and stepped up in the fight, didn't act like a coward at all, and managed to win the fight along side Chopper.
A clear pattern shows in all of Usopp's fights, where when around the people that he looks up to, he acts like a coward to try and get them to help him, but when he is with Chopper or just alone, he can't act like a coward or they will lose the fight. Through the entire story, everytime Usopp shows actual true fear, is when fear is warranted, all of the other times he is faking it, which is why it is so over the top.
In Water 7, Usopp gets mad at Luffy and in his anger, forgets about acting like a coward and fights Luffy as himself. AND HE DOES MORE DAMAGE TO LUFFY THAN ENEL DID. He then goes on to create Sogeking, the epitome of what his goal in life is. He tells everyone that he isn't Usopp because in his mind, if he's not Usopp, he doesn't have to act like a coward. And he fights at the top of his game the entire arc.
Finally, in Thriller Bark he fight Perona alone. During the fight he puts his mask back on to become Sogeking because he thinks that Sogeking is a better fighter than he is, but having to act like a coward is ingrained so much into his mind that he doesn't realize that if he didn't pretend to be a coward all the time, he would be one of the best fighters on the crew.
