I'm looking for an anime to cheer me up since I've been feeling pretty down lately. It doesn't necessarily have to be completely happy or lighthearted, but an uplifting, fun or amusing plot with interesting characters would be great. Beautiful art and animation make me happy too!

I just checked your MAL list and saw that you have not watched Working! yet. This was recommended to me when I started to watch a lot of Anime and it was funny. ( IMO) and a lot of other peoples. There is no Echii which is a welcome relief and the story is very good. There are 2 seasons to the series so if you like the first and that manages to cheer you up a little.

If you watch the second season you will have a smile on your face...

Hope you get to enjoy the seires as much as I did.

The Aria series (starting with Animation, not Origination!) is a very happy, uplifting anime. It also has some beautiful art.

Whisper of the Heart, Bamboo Blade, and Minami-ke are also all very good slice-of-life series that are funny and sweet.

Seconding Aria
in similar spirit Tamayura, Azumanga Daioh or Hidamari Sketch (last two are comedies but good ones)

Kimi ni Todoke for romance that has same feel to it

if you want something more lively than Kaleido Star - it may be longer but very warm (even if/because of there is drama here and there) and with lovelly characters

Nichijou - Comedy, School, Slice of Life

Seto no Hanayome - Comedy, Parody, Romance, School

Seitokai no Ichizon - Comedy, Parody, School, Harem

Inu x Boku SS - Comedy, Shounen, Supernatural

K-On! - Comedy, Music, School, Slice of Life

Hanamaru Youchien - Comedy, Slice of Life

some comedy animes like: danshi koukousei no nichijou or level E (gintama is great too, try re watching some good episodes, it works for me :p )
or some light animes like: kimi to boku, usagi drop, natsume yuujinchou... i recommend working !! & k-on too

Really, I think the best recommendation is Working; it is silly and cheerful and energetic, and it is not girls only like most anime of its kind. Second season is much better than the first.

Other anime proposed are also very good, but many are simply relaxing without being too energetic. I think the best thing to dispel the blues is genki cheerfulness.

I'm totally going to second Kimi to Boku, Usagi Drop, and Natsume. I feel like an idiot for forgetting these!

Actually I found the GotF cheerful and heartwarming. It was only when it starts to approach the ending when the mood changes

I'm the third one to say this but I do recommend Ouran Host Club, I watch it all the time and read the manga. I watch it when I'm down, feeling sick and when I'm happy too!

I'd also recommend Usagi Drop which is something I watched recently, it's a really cute slice of life anime involving an orphaned girl who's adopted by a family member who has no idea what he's doing. There are some sad bits, but it's more of a cute sad and it's really enjoyable to watch.

Then the latest thing I watched was Kaichou wa Maid-sama!, it's funny with some romance thrown in. It's great if you like an independent strong female lead who realises she can rely on other's because even though she's stronger than most of the men around her, sometimes she meets her match but it's okay to ask for help.

[I should like I'm getting paid to advertise some of these shows. Hah! Hope my suggestions helped.]
