There are many proto-tsundere, but the one that defined the trope as a stock character, imo, is Asuka. It's still not as flat as most of modern tsundere, but think they're all modeled after her. Just like all kuudere are Rei's copies.

Though the term tsundere hadn't been coined yet, the earliest Tsundere in anime that I know, is Mai Ookubo from Igano Kabamaru (1983).
I haven't seen Urusei Yatsura but I'm not sure that Lum should count as a Tsundere.

The earliest tsundere is of course Hera from Greek mythology (circa 900 B.C.E.). She's always a bitch to Zeus but you know she cares about him even if he goes to flirt with the mortals.

i dont count lum as a tsundere because she only gets angry when ataru is being an idiot.

I am 100% certain the first tsundere was Lum which got us the more refined character in Tendou Akane a few years later from Takahashi Rumiko-sensei. To me, Akane is the first real Tsundere, and one of the very few really likable ones from that character type. You cannot blame her to be tsuntsun seeing her fiance like that! She genuinely thought that she should be the only person worthy of Ranma's affections because she was his fiancee. And she would even point out that from time to time. She would get mad at him but would still wait for him to come back to her and was genuinely kind.

We see so many modern day Tsundere characters but none can surpass Akane. Most of them are not very likable to begin with.
