They are both balls to the walls insane when it comes to comedy. But Bobobo-bo Bo-bobo is more of a full randomness, 100Kanojo is a harem Romcom with full of parodies and bunch of shameless references with a dash of wholesome.

Both are full of meta-humor 4th wall break jokes, and even interact with the author itself. Comedy is the same even with the execution of the jokes. The difference is that while 100 kanojo is more of a romcom SoL seinen, Gintama is a full shonen with surreal jokes, rich and simple plot, and it's mixture of being an absurd yet light-hearted comedy and a serious yet engaging action-drama.
