So I saw some of the kind and even though fun (Kampfer for example) I was wondering if there was an anime with a more "serious" tone to it. Normally the main character is already girlish before the transformation, and isn't very concerned about it. It ends up being a yuri-fest (not that I don'r like :3 ). A character that actualy acts like a guy, and then transforms would be a funny setting.

If you still want comedy, but with a genderbend that acts like a guy most of the time, go with Ranma.

May not be up your alley, but thought I'd throw out there the fact that the final season of Sailormoon (StarS) features prominently three characters who transform back and forth between genders. Although the show itself isn't always serious, the fact that they are genderbending is never comedic.
