So first things first, I haven't read the manga yet, so I'd appreciate it if the answer to my question could be summarized based on what has been shown in the anime (as of ep 2).

Early into the episode, when Miyako and Ruby are talking about starting an idol group etc., Miyako starts a sentence by saying 'Besides..' before stopping. Looking back at this part, her surprise when Ruby said she was scouted and her conversation with Aqua later in the episode (regarding the stuff the idol from the underground says and him hiring a detective etc.), I have been wondering if Miyako had already known about Aqua's plan to make the fake call.

No, she didn’t know his plan until afterwards. It’s implied in the manga that she doesn’t know it in the scene you are talking about, but realizes it sometime afterwards, in between that conversation and the actual phone call.
