Does one need to be familiar with the series to be able to enjoy this film?

No. This is an alternative version of the first Macross anime. There is a prequel, but you don't have to see it in order to understand and enjoy this movie.

Yes... if only because things happen a bit too fast in the film and you will not be able to grasp the emotional situation of the characters and appreciate the differences without knowing them beforehand... though I guess you could enjoy it even as a standalone, without knowledge of the original series you'd miss out a lot.

You can pretty much watch the first 4-5 episodes of the Macross TV series to fully understand the needed backstory for the movie. But then the some of the character relations and other things change, but it's not too bewildering to understand.

It's perfectly understandable without seeing the T.V show.

However how much of the enjoyment will be lost is a really good question.
A lot of the humour and more subtle moments will be complete blank spots for those who haven't seen the show.

In order to fully enjoy this movie you must watch the TV series first. As others have said, this movie is perfectly understandable as a standalone but not nearly as enjoyable.

No - this is actually the original before any of the series were created. Everything else is based, either directly or indirectly, on this movie.

Not true - while the series do fill in different aspects, there is no need to watch them before the movie.

This was made to be watched as a sequel, not as a stand alone film, since most of what happens here can only resonate if you already know the characters or what's at stake, and besides, watching this first will only spoil truly emotional moments from the original series that in this particular film mean nothing. There's no proper buildup for many huge moments, many poorly explained or outright unexplained things (like, how far into the future are we? why is a city inside this warship? why are humans fighting aliens so effectively? is Earth the only populated planet? are there more Macross-style vessels out there?) and the amount of implausible stuff happens so fast, that the viewer will end up feeling dumbfounded.

So beware those who pretend to watch this first, it may be tempting but it's not the proper way to start with this franchise.
